Sluggy Freelance http://www.sluggy.com (One of the longest if not the longest webcomics still running, updates daily, mostly only new content on week days anymore though. genre: little bit of everything)
Sluggy Freelance is hard to explain. It starts out as mostly parody of popular fandom stuff, but soon comes into it's own. Some of the more recent plotlines are pretty creative. My only complaint is that the character development isn't great. Of course, you could easily argue that this comic would be worse off if it was =P

Misfile - http://www.misfile.com (updates weekdays, genre: supernatural, transgender, cars)
This one's kind of unique. Just read it, it's one of my favorites. I tried writing a description, but I can't seem to write one that's adequate here.

Girl Genius http://www.girlgeniusonline.com/comic.php (updates MWF, genre: steampunk)
Besides being the only steampunk webcomic I've ever seen, it's a pretty magnificent example of storytelling. Besides, mad science always makes for an interesting plot.

Flipside http://www.flipsidecomics.com/comic.php  (updates MWF usually, not always. Genre: Fantasy, humor, drama)
It's a fantasy/magical setting, but it's not the conventional rip-off of certain popular author's worlds that so many other fantasy/magical comics are. It's got a more modern feel to it, but more importantly, the author is good at telling a decent story in his own style.

Those are a few of the major ones I read that aren't already on the list. Frankly I'm shocked Sluggy Freelance isn't on there.

Edit: will post more in a bit.

Some more:

It's Walky! / Joyce and Walky / Roomies! http://www.joyceandwalky.com/
This one's hard to explain. It's got three... stages of sorts. They're not plot arcs, it's more than that, you'll have to read it to understand. The current comics are, except for a few about two of the characters lives, unfortunately pay-only (although it's pretty cheap), none the less there's a MASSIVE archive to read through and you won't be left hanging, the pay part is a separate arc of sorts. It's one of my favorites. And be warned, it's freaking LONG. Not as long as sluggy freelance, perhaps, but still, one of the longest I've read yet. Reccomend this one to everyone. Oh, the first and second parts are different genres. The first could be considered slice-of-life/romance/college/minor fantasy/etc, the second is more sci-fi/romance.

Shortpacked! http://www.shortpacked.com/ - Same author, and has some of the characters from the above. Technically same world, but genre is very different. I kind of liked it despite having nothing really in common with the target audience, your mileage will vary.

College Roomies From Hell -http://www.crfh.net/ This one is kind of hard. The drawing at first sucks. Ok, the art at the start REALLY sucks, and the plot isn't much better. It slowly gets better though, and by the point it's at now it's much better, although it doesn't update as often anymore. I can't reccomend this one to everyone, but it's worth a try. I do warn you, it's pretty long, up there with Walky and Sluggy Freelance in length.

Ghastly's Ghastly Comic http://www.ghastlycomic.com/ - Warning: This comic has the potential to offend a lot of people in really unpleasant ways. It's not that it TRIES to offensive so much as it... It is what it is. The comic doesn't really update anymore, but there's a rich archive. Also, the art dramatically improves about half-way through if you don't like the way it looks initially.

Sexy Losers http://www.sexylosers.com/ - Like the above, has the potential to offend, although the style is decidedly different. Also like the above, it isn't offensive for the sake of being offensive, like South Park and other various comedies are. The comic is potentially offensive simply due to the nature of the content.

Edit: Missed that this one was already on the main page
Another vote for Perry Bible Fellowship:
This has to be the most surreal comic I've every seen. It's disturbing in its simple yet terrifying suggestions and art style.

I'm going to edit my previous post to include better descriptions as well, and then I'm going to scour my now out of date archives for other ones I probably missed.

Just as a personal comment... While I certainly haven't read all those on the list right now, "Least I could do" I've read, and while it's not the worst webcomic I've ever read, it's pretty far down there. The art isn't very good, and more importantly, neither are the jokes. Also, and I know I'll get flamed for this... Cyanide and Happiness isn't that good. Yeah, the earlier material is funny, but the later stuff is just... bland. Almost lame really. If you want a great stick figure comic, look no further than XKCD (already on the list).

Of those on the list that I have read, most are actually pretty good. And believe me, I've read a lot of webcomics.

Edit: Another one.

Errant Story http://www.errantstory.com/ When I first starting reading this, I thought it going to be another crappy fantasy rip-off. I was pretty wrong. Let's just say that things aren't exactly what they seem at first.

I actually happen to enjoy the daily updates of LICD. Sure, the art doesn't make you jump in the air with joy, but it's still pretty decent. And sure, the jokes are a bit repetitive, but they work in relation to the characters, and add some definition to it all. Also, they have some well thought out story arcs, that keep reading exciting. The site also features animated shorts, which I think is pretty snazzy.

Sure, it's not as rugged as some guy sitting in his (mother's) basement doodling, as Sohmer actually runs an animation company through which he produces LICD, as well as Looking For Group, amongst other projects. But I still think it's a good comic, which is reflected in it's book sales.

Just because I started this thread doesn't mean I know all the cool shit out there. Really, this is just for me to find more cool shit. Hence why ASW is now with my other RSS feeds.

I would like to make a sincere apology. Somehow, somewhere along the line, my brain mixed up "Least I could do" with another comic whose name escapes me now.

I've since gone back and looked at the real comic, and while I still have issues with it, it's not nearly as bad as the one I was thinking of when I wrote my post.

Very sorry about that.

Apology accepted.

Interesting note: LICD was nominated for a WCCA last year.
