Depends on what you'll use the picture for.
If you want lossless quality then use PNG or if you want small size use JPEG.

PNG is good for quality, JPEG is good for size. Avatars for LW I either always do in JPEG or GIF because of size. Avatars for elsewhere or other images I may edit in Photoshop, I save as PNG.

PNG is lossless and it also has support for alpha transparency. For web graphics I liek to use PNg because of that, however you have to rememebr that IE 6 doesn't support PNG transparency.

JPEG is better for photos because although it is lossy, the compression is much better and it is designed to take advantage of the way the human brian sees an image so that the data discarded is not so noticable. Certainly with large files I'd use JPEG. The difference even between a 100% quality JPEG and a 75% quality is not every noticable in most conditions but can represent as much as 60% compression.

So I say PNG for graphics and JPEG for "natural" or large images.
