No. That's absolutely ridiculous.  

You should only be worried about moles if they change shape, colour, are bigger in diameter than a pencil eraser or if the edges are bumpy or jagged instead of smooth.

However, if you don't protect your skin in the sun, you should get your moles checked once a year by a dermatologist, just in case.

That would have to be irritation for a very, very long time. If you scratch it pretty much non-stop for months the plasticity of the cells would change dramatically and yes, a precursor to cancer might form.
